Analysis of inflow sources of free-living amoebas in a Korean river

Abstract
Multiplex nested PCR for detection of Acanthamoeba spp. and Naegleria fowleri was conducted on samples collected from bioreactor sludges and effluents of sewage treatment plants (STPs) and river samples in CJ City to reveal the inflow source of pathogenic free-living amoebas (FLAs) found in Korean rivers. The sequence diversity of the 18S rRNA gene of Naegleria spp. was also analyzed. 18S rRNA gene of Acanthamoeba spp. was detected in all samples collected in this study. As a result of analyzing the sequences of the 18S rRNA gene of Naegleria spp. in river and effluent samples, 24.3% of the sequences were more than 99.5% similar to N. italica or N. australiensis, which are known to cause diseases in animals. In addition, the composition of sequences of Naegleria spp. in the effluent of the STP and the river sample collected directly downstream of the discharge point were very similar, suggesting that the STP is one of the inflow sources of Naegleria spp. found in rivers.
